چکیده

A monoclonal antibody (mAb) that recognizes the y chain of human fetal hemoglobin(Hb F) has been produced by cell hybridization techniques. The mAb reacts with Hb F (a2y2), Hb Bail’s(y),and Hb Kenya (yhybrid), butdoes notcrossreactwith Hb A (a) or Hb A2 (a2&). We describe a direct enzyme-linked immunoassay (ELISA)for measurement of Hb F, in which hemoglobins from standards or from unknown hemolysatesare covalentlyboundto the wells of microtiter plates. The antigen is quantified by addition of the 7-specific mAb, followed by anti-mouse IgG conjugated with horseradish peroxidase, and incubationwith the substrate, tetramethylbenzldine. Absorbances at630 nm aredirectly proportional to the amount of Hb F present in the standards or samples. Results for Hb F in 53 hemolysates agreed well withvalues obtainedby “high-performance”liquidchromatography,RIA, alkali denaturation, and magnetic affinity immunoassay. This EUSA can detect a 0.5% proportion of Hb F in 1 h and offers distinct advantages over othertechniquescurrentlyin use.

The Assessment of Feto-Maternal Hemorrhage in an Artificial Model Using Anti-D and Anti-Fetal Hemoglobin Antibody by Flow Cytometry

Background: When fetal red cells enter the maternal circulation from placenta, an event would be happened that is described as feto-maternal hemorrhage (FMH). This life-threatening condition could be detected by using RBC antigens (surface antigens and intracellular antigens). Therefore, the measurement of fetal RBC in an artificial model would be useful to calculate FMH and consequently the do...

Measurement of Affinity Constant of Anti-human IgG Monoclonal Antibodies by an ELISA-based Method

Background: The affinity of an antibody to its antigen is a crucial parameter in its biological activity and performance of an immunoassay such as ELISA. Affinity of most IgG specific MAbs are often determined by methods which require labeling of either antigen or antibody, and are sometimes difficult to control, do not always lead to the expected signal and often result in immunological modifi...

Monoclonal and Polyclonal Antibodies Specific to Human Fibromodulin

Background: The unique expression of fibromodulin (FMOD) in patients with chronic lymphocytic leukemia (CLL) has been previously reported. Detecting FMOD in CLL patients using specific anti-FMOD mAbs might provide a promising method in detection, monitoring, and prognosis of CLL. Objectives: In this study, we aimed for producing specific antibodies agains...
